DNA sexing using a dried blood spot on paper involves extracting DNA from the blood sample collected on the paper. The process begins by punching out a small piece of the blood spot card containing the dried blood.

Next, the DNA is isolated from the blood sample using various extraction methods to purify the genetic material. Once the DNA is extracted, specific genetic markers on the sex chromosomes, like the amelogenin gene, are amplified using Polymerase Chain Reaction (PCR). Overall, DNA sexing from a dried blood spot on paper is a reliable and efficient method for determining the sex of an individual for various applications, such as gender determination in newborn screening or wildlife conservation.
