Introduction:
DNA sex testing through blood spot samples is a reliable and convenient method for determining the sex of birds. This guide will provide step-by-step instructions on how to take a DNA blood spot sample for DNA sex testing.
Materials Needed:
- Ziplock bag
- Disposable gloves
- Swabs from Amazon: Please only use these!
Procedure:
- Prepare Your Work Area:
- Choose a clean and well-lit area for sampling.
- Lay out all necessary materials within reach.
- Prepare the Bird:
- Handle the bird gently and securely to minimize stress.
- If the bird is particularly nervous or wriggly, you may want to enlist the help of a second person to hold it securely during sampling.
- Collect the Swab Sample:
- For Buccal (Mouth) Swab:
- Gently restrain the bird to minimize stress.
- Insert the sterile swab into the bird’s mouth, ensuring to swab the inner cheek by rotating the swab firmly against the cheek lining for 5-10 seconds.
- For Cloacal (Vent) Swab:
- Gently restrain the bird.
- Carefully insert the sterile swab into the cloaca about 1-2 cm and rotate gently for 5-10 seconds.
- Drying the Swab:
- Allow the swab to air dry completely for approximately 10-15 minutes. This step is crucial to prevent mold growth during transit. Please ensure the sample is in it’s own separate bag.
- Packaging the Sample:
- Once dry, place the swab into a paper envelope. Avoid using plastic bags as they can retain moisture.
- Seal the envelope securely
- Use a permanent marker to clearly write the bird’s identification details (e.g., species, age, sex) and your contact information on the envelope.
- Send the Sample for Testing:
- Send the package to the designated address using your preferred shipping method.
