How to Take an Eggshell Sample for DNA Sex Testing

Introduction:

DNA sex testing through eggshell samples has revolutionized avian breeding and conservation efforts, providing accurate and non-invasive methods for determining the sex of birds. This guide will walk you through the process of taking a DNA feather sample for DNA sex testing.

Materials Needed:

  1. Alcohol wipes or rubbing alcohol
  2. Disposable gloves (optional but recommended)
  3. Small zip-lock bag

Procedure:

  1. Prepare Ahead of Time:
    • With this type of testing, you need to plan to do this before your eggs hatch.
    • Separate the eggs with dividers of your choosing when you set them for lockdown.
  2. Put a band or some type of identifier on each chick as you pull it out of the incubator.
  3. As you pull out each chick, label a ziplock bag with the same Bird ID and set the eggshell next to it or on it.
  4. Ensure the eggshell has fully dried out, and gently flatten it so it will be easier to ship (you can also leave it whole if you chose to ship in a box). Please ensure that there is dried membrane inside the eggshell, as that is what we need to test.
  5. Once the eggshell has fully dried it out, please put it in it’s labeled ziplock bag and seal it.
  6. Secure the Sample:
    • Seal the packaging securely to prevent contamination or loss of the sample. Do not use the same Bird ID for multiple birds for orders you are sending in at the same time.
    • Please ensure the sample is in it’s own separate bag.
  7. Dispose of Waste:
    • Dispose of used gloves (if applicable) and any other waste materials properly.
    • Clean your work area and tools with alcohol wipes or rubbing alcohol to ensure they are sterile for future use.
  8. Send the Sample for Testing:
    • Staple a piece of paper to the bag with the Bird ID you listed on the website. Or label the bag itself.
    • Send the package to the designated address using your preferred shipping method.
