How to Take a Feather Sample for DNA Sex Testing

Introduction:

DNA sex testing through feather samples has revolutionized avian breeding and conservation efforts, providing accurate and non-invasive methods for determining the sex of birds. This guide will walk you through the process of taking a DNA feather sample for DNA sex testing.

Silkies must be at least four weeks old or older to Feather Test.

Materials Needed:

  1. Clean tweezers
  2. Alcohol wipes or rubbing alcohol
  3. Disposable gloves (optional but recommended)
  4. Small zip-lock bag

Procedure:

  1. Prepare Your Work Area:
    • Choose a well-lit, clean, and quiet area to minimize stress on the bird during sampling.
    • Lay out all necessary materials within reach.
  2. Prepare the Bird:
    • Handle the bird gently and securely to minimize stress.
    • If the bird is particularly feisty, you may want to enlist the help of a second person to hold it securely during sampling.
  3. Identify the collection area:
    • Select a few clean and undamaged feathers from the bird’s chest area. The feather should be plucked rather than cut to ensure an intact DNA sample. Please pluck 3-4 feathers at least.
    • Avoid feathers with dirt, debris, or signs of damage.
  4. Sterilize Tools and Hands:
    • Sterilize tools by wiping with alcohol.
    • If not using gloves, thoroughly clean your hands with soap and water or use disposable gloves.
  5. Collect the Feather Sample:
    • Gently pluck the selected feather from the bird, ensuring minimal discomfort.
    • Hold the feather close to the base and pluck it swiftly and firmly to minimize stress on the bird.
    • If using tweezers, grasp the base of the feather close to the skin and pull gently to remove it.
    • Place the plucked feather on a clean surface.
    • The feather sample should look similar to the photo at the bottom of the page – look closely at the “tip” of the feather; that is the area we actually use for testing.
  6. Secure the Sample:
    • Put all plucked feathers from each bird into their own bags. 
    • Seal the packaging securely to prevent contamination or loss of the sample. Do not use the same Bird ID for multiple birds for orders you are sending in at the same time.
    • Please ensure the sample is in it’s own separate bag.
  7. Dispose of Waste:
    • Dispose of used gloves (if applicable) and any other waste materials properly.
    • Clean your work area and tools with alcohol wipes or rubbing alcohol to ensure they are sterile for future use.
  8. Send the Sample for Testing:
    • Staple a piece of paper to the bag with the Bird ID you listed on the website.
    • Send the package to the designated address using your preferred shipping method.
